چکیده

The circumventricular organs (CVOs) are structures bordering the third and fourth ventricles and are considered to be important sites for communicating between neural cells, cerebrospinal fluid and blood. CVOs have been characterized as regions of the brain with permeable capillaries, which can pass marker dyes from blood (Davson et al. 1987). This high tracer transportability of vessels in the CVOs was explained by the histological findings of the capillary endothelium, which has cytoplasmic vesicles, caveolae, pits, fenestrations and a few tight junctions (Bouchaud & Bosler, 1986; Gross, 1992). Several substances such as horseradish peroxidase and aaminoisobutyric acid (AIB) have been used as markers, and it has been revealed that the various parts of the CVOs have different permeability values. The order of permeability of AIB from highest to lowest was pineal gland > the posterior lobe of the pituitary gland > the subfornical organ (SFO), area postrema (AO), and median eminence (ME) > subcommissural organ (SCO) (Brodie et al. 1960a,b; Broadwell & Brightman, 1976; Bouchaud & Bosler, 1986; Gross et al. 1987; Gross, 1992). The SFO might be a part of the sensory system involved in body fluid homeostasis, and it might transfer information on the osmolarity of serum to neurons. However, no direct observations on water permeability in the SFO have been reported.

Water permeability of capillaries in the subfornical organ of rats determined by Gd-DTPA(2-) enhanced 1H magnetic resonance imaging.

The water permeability of capillaries in the subfornical organ (SFO) of rat was measured by a (1)H nuclear magnetic resonance method in combination with a venous injection of a relaxation reagent, gadolinium-diethylene triamine-N,N,N',N",N"-pentaacetic acid (Gd-DTPA(2-)), which could not pass through the blood-brain barrier (BBB). Judging from results of Gd-DTPA(2-) dose dependency in the intac...

Gadolinium-Diethylenetriaminepenta-Acetic acid Conjugated with Monoclonal Antibody C595 as New Magnetic Resonance Imaging Contrast Agents for Breast Cancer (MCF-7) Detection

Background: The monoclonal antibody, C595, against breast cancer cell line was conjugated with cyclic anhydride gadolinium-diethylenetriaminepenta-acetic acid (Gd-cDTPAa) to produce Gd-DTPA-C595 and used as specific breast cancer cell line (MCF-7) contrast agents in magnetic resonance imaging (MRI).  Methods: After incubation of breast cancer cell line (MCF-7), with different contrast agents (G...

Simultaneous MRI measurement of blood flow, blood volume, and capillary permeability in mammary tumors using two different contrast agents.

A technique for the simultaneous measurement of three vascular parameters: blood flow (Frho), blood volume (v(b)), and the capillary permeability-surface area product (PSrho) in breast tumors using dynamic contrast-enhanced magnetic resonance imaging (MRI) is presented. Features of the technique include measurement of precontrast tumor T(1), rapid temporal sampling, measurement of the arterial ...

Dynamic Contrast Enhanced MRI (DCE-MRI): From the Simplest to the Most Advanced DCE-Model

1. Tracer Kinetic Modeling • Extracellular agents (such as Gd-DTPA) accumulate in the interstitium of many tissues (e.g. muscle) but do not cross the healthy blood-brain-barrier. • Functional information may be obtained by treating Gd-DTPA as a tracer and following its kinetics (as opposed to the conventional ‘static’ approach, inject wait – image once). • After intravenous administration (norm...

Dynamic contrast-enhanced MRI measurements of passive permeability through blood retinal barrier in diabetic rats.

PURPOSE To test the hypothesis that dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) provides a useful in vivo measure of passive blood retinal barrier permeability surface area product (BRB PS) in experimental diabetic retinopathy. METHODS BRB PS (cm(3)/min) was measured using DCE-MRI and Gd-DTPA (MW 590 Da) in urethane-anesthetized control rats, sodium iodate-treated rats, rat...
